Project Abstract

This study undertakes a comprehensive comparative analysis of phytochemical profiles and antimicrobial activities of endophytic fungi isolated from selected medicinal plants in [region], aiming to elucidate the chemical diversity and therapeutic potential harbored within plant-associated fungal communities. Through systematic isolation from healthy plant tissues, a diverse collection of endophytic fungi was obtained and purified, followed by species-level identification using morphological characterization and molecular barcoding of the ITS region. Quantitative and qualitative phytochemical screening was performed on fungal extracts using chromatographic (HPLC-DAD, LC-MS/MS) and spectroscopic methods to map the repertoire of secondary metabolites, focusing on classes with known antimicrobial relevance such as alkaloids, terpenoids, polyketides, and phenolics. Multivariate chemometric analyses, including principal component analysis and hierarchical clustering, were employed to compare phytochemical fingerprints across fungal taxa and plant hosts, revealing host-driven and tissue-specific metabolite signatures. Antimicrobial assays were conducted against a panel of clinically relevant pathogens, including multidrug-resistant bacteria (e.g., MRSA, Pseudomonas aeruginosa) and opportunistic fungi (e.g., Candida auris), using broth microdilution to determine minimum inhibitory concentrations (MICs) and minimum fungicidal concentrations (MFCs). Bioassay-guided fractionation identified bioactive constituents, and synergy assessments (checkerboard and time-kill kinetics) evaluated potential combinatorial effects with standard antibiotics. The results demonstrate substantial inter- and intra-species variation in metabolite production, with several endophytes producing novel or scarcely reported compounds exhibiting potent antimicrobial activity, including activity against resistant strains. Notably, extracts from endophytic fungi isolated from traditionally used medicinal plants displayed higher chemical diversity and stronger bioactivity on average, suggesting a correlation between ethnobotanical value and endophytic metabolic potential. Mechanistic insights were explored through preliminary target-based assays, indicating possible disruption of bacterial cell wall synthesis, protein biosynthesis, and membrane integrity, as well as fungal ergosterol pathway interference. Genomic and transcriptomic analyses of promising isolates are proposed to identify biosynthetic gene clusters responsible for metabolite production and to assess regulatory factors governing antimicrobialCompound biosynthesis under plant-derived cues. The study contributes to the understanding of the ecological role of endophytic fungi in medicinal plants, highlights the untapped reservoir of natural products with antimicrobial properties, and identifies candidate strains and compounds for further preclinical development. The integration of phytochemical profiling with bioactivity data provides a robust framework for prioritizing endophyte-derived metabolites for drug discovery, while emphasizing the influence of host plant selection and tissue type on endophytic chemistry. Limitations include potential biases from culture-dependent isolation, the need for dereplication to avoid redundancy, and the necessity for scalable production of promising metabolites. Overall, the findings advance the field of endophytic fungal chemistry and offer a foundation for future translational research targeting antimicrobial resistance.
