Project Abstract

The emergence of multidrug-resistant (MDR) pathogens poses a significant threat to global health, compromising the effectiveness of conventional antibiotics and leading to increased morbidity, mortality, and healthcare costs. In the face of this growing challenge, the exploration of alternative antimicrobial agents, particularly from natural sources, has become a crucial area of research. This project aims to investigate the antimicrobial potential of various natural compounds against a range of MDR pathogens, with the ultimate goal of identifying promising candidates for the development of novel therapeutic interventions. The project will focus on a comprehensive screening and evaluation of a diverse array of natural compounds, including plant extracts, essential oils, and isolated phytochemicals, for their ability to inhibit the growth and proliferation of selected MDR bacterial and fungal strains. These target pathogens will be carefully selected based on their clinical relevance, prevalence, and resistance profiles, encompassing both Gram-positive and Gram-negative bacteria, as well as fungal species known to cause serious infections in immunocompromised individuals. The experimental approach will involve a multifaceted methodology, including in vitro antimicrobial susceptibility testing, bioactive compound identification and characterization, and mechanism of action studies. The antimicrobial activity of the natural compounds will be evaluated using standard microbiological techniques, such as broth dilution and agar diffusion assays, to determine their minimum inhibitory concentrations (MICs) and minimum bactericidal/fungicidal concentrations (MBCs/MFCs). Additionally, the project will explore the synergistic effects of combining natural compounds with conventional antibiotics, with the potential to enhance the overall antimicrobial efficacy and overcome resistance mechanisms. To gain a deeper understanding of the underlying antimicrobial mechanisms, the project will employ advanced analytical techniques, including high-performance liquid chromatography (HPLC), mass spectrometry (MS), and nuclear magnetic resonance (NMR) spectroscopy, to identify and characterize the active phytochemical constituents responsible for the observed antimicrobial activities. Furthermore, the project will delve into the elucidation of the mode of action of the most promising natural compounds, investigating their effects on various cellular processes, such as membrane integrity, metabolic pathways, and virulence factor production, using a combination of microscopic, biochemical, and molecular biology approaches. The findings of this project are expected to contribute significantly to the understanding of the antimicrobial potential of natural compounds and their ability to combat MDR pathogens. The identification of potent and selective natural antimicrobial agents, as well as the insights into their mechanisms of action, will provide valuable information for the development of innovative therapeutic strategies and the potential for their integration into existing antimicrobial treatment regimens. Moreover, the project's outcomes may lead to the discovery of novel lead compounds or synergistic combinations that could inspire the design of future antimicrobial drug development efforts, ultimately contributing to the global fight against the growing threat of antibiotic resistance.
